- Seeds contain 5-hydroxy-L-tryptophan, or 5-HTP, a serotonin precursor. Researchers developed a direct high-performance liquid chromatography assay. The presence of a molecule and the ability to measure it do not establish therapeutic benefits from the whole plant.

What clinical research can establish

- Javelle and colleagues’ review included thirteen 5-HTP studies, seven in the quantitative analysis. Despite favourable signals, it highlighted substantial heterogeneity, generally weak quality and few placebo groups. It does not establish Griffonia as an effective antidepressant, sleep medicine or weight-loss product.

- A historical Cochrane review retained only two trials, totalling 64 participants, for an analysis combining tryptophan and 5-HTP. It found the evidence insufficient for firm conclusions. Its age and pooling of substances must remain visible: it is not a comprehensive review of current research.

Laboratory study
- A 2019 study of Ivorian-origin seeds measured about 156 mg of 5-HTP per dry gram, alongside other alkaloids and fatty acids. It combined chromatography and DNA fingerprinting for authentication. This sample result does not predict the local plant’s concentration and is not equivalent to a purified extract’s percentage.
Dry seed mass, chemical fraction and concentrated extract have different denominators.

Traditional account
- A 2016 survey in Ejisu-Juaben, Ghana, lists Griffonia simplicifolia as Kagya and records seed use for stomachaches. A voucher specimen is cited: CPMR 4073. This is a local ethnobotanical report.

A lectin studied against insects
Laboratory study
- GS-II lectin recognises N-acetylglucosamine. Experiments on the cowpea weevil Callosobruchus maculatus link its insecticidal activity to carbohydrate binding and resistance to protein digestion. A purified laboratory protein is not a validated homemade pesticide.
